GENE MUTATION PATTERNS OF RIFAMPICIN IN MULTIDRUG RESISTANT MYCOBACTERIUM TUBERCULOSIS COMPLEX STRAINS

Objective: The aim of this study was to evaluate the GenoType®MTBDRplus test for detection rifampicin (RIF) resistance in MDR Mycobacterium tuberculosis complex strains. Materials and Methods: Twenty-five multidrug-resistant (MDR) clinical strains were used, gene mutations causing RIF investigated by results compared with BACTEC 460 TB system. strain sequenced if there no mutation absence Wild Type (WT). ATCC 35838 used as a quality control (QC) strain. Results: There 96% compliance between system finding resistance. most frequent zone rpoB S531L promotor (13 strains, 52%). other H526Y (three 12%) H526D 12%), while five (20%) had Δ2-Δ5 wild type probes. only one (4%) but it found be resistant This detected have triple mutations. Mutations on codons 489, 493, 503. Conclusion: showed good compatibility detecting resistance, thought that could an effective reliable susceptibility testing MDRTB patients, providing significant advantage technical time.
