چکیده
Objective: The aim of this study was to evaluate the GenoType®MTBDRplus test for detection rifampicin (RIF) resistance in MDR Mycobacterium tuberculosis complex strains. Materials and Methods: Twenty-five multidrug-resistant (MDR) clinical strains were used, gene mutations causing RIF investigated by results compared with BACTEC 460 TB system. strain sequenced if there no mutation absence Wild Type (WT). ATCC 35838 used as a quality control (QC) strain. Results: There 96% compliance between system finding resistance. most frequent zone rpoB S531L promotor (13 strains, 52%). other H526Y (three 12%) H526D 12%), while five (20%) had Δ2-Δ5 wild type probes. only one (4%) but it found be resistant This detected have triple mutations. Mutations on codons 489, 493, 503. Conclusion: showed good compatibility detecting resistance, thought that could an effective reliable susceptibility testing MDRTB patients, providing significant advantage technical time.

Mutations in the RpoB Gene of Multidrug-Resistant Mycobacterium Tuberculosis Isolates from Semarang, Indonesia
ABSTRACTTuberculosis is an infectious disease which occurs widely in the world and becomes one of the top 10 causes of death worldwide. Mutations of the RpoB gene cause the resistance of Mycobacterium tuberculosis to rifampicin that contributes to the occurrence of MDR-TB. This study aimed to determine the pattern of rpoB gene polymorphisms in the MDR M. tuberculosis in Semarang, Indonesia. Mos...

Multidrug-resistant (MDR) Mycobacterium tuberculosis complex (MTBC) strains represent a major threat for tuberculosis (TB) control. Treatment of MDR-TB patients is long and less effective, resulting in a significant number of treatment failures. The development of further resistances leads to extensively drug-resistant (XDR) variants. However, data on the individual reasons for treatment failur...

In 2013 alone, the death rate among the 9.0 million people infected with Mycobacterium tuberculosis (TB) worldwide was around 14%, which is unacceptably high. An empiric treatment of patients infected with TB or drug-resistant Mycobacterium tuberculosis (MDR-TB) strain can also result in the spread of MDR-TB. The diagnostic tools which are rapid, reliable, and have simple experimental protocols...
متن کاملMolecular characterisation of rifampicin-resistant Mycobacterium tuberculosis strains from Malawi
BACKGROUND Availability and access to the detection of resistance to anti-tuberculosis drugs remains a significant challenge in Malawi due to limited diagnostic services. The Xpert® MTB/RIF can detect Mycobacterium tuberculosis and resistance to rifampicin in a single, rapid assay. Rifampicin-resistant M. tuberculosis has not been well studied in Malawi. OBJECTIVES We aimed to determine mutat...

The surveillance of drug resistance, using mycobacterial cultures and in vitro susceptibility tests, allows the estimation of the regional and worldwide magnitude of this phenomenon in tuberculosis. This study, which is part of 3 international projects organized by WHO/UICTER, determined the prevalence of the resistance to antituberculosis drugs in Cuba, during the 1995-2000 period, in both naï...
